Brigatinib is used to treat a specific type of non-small cell lung cancer (NSCLC) that has metastasized to other parts of the body. It belongs to a class of medications called kinase inhibitors, working by blocking the action of an abnormal protein that signals cancer cells to multiply, thus slowing or halting the spread of cancer cells.

  • Brigatinib is administered orally as a tablet, usually once daily, with or without food.
  • Take it at the same time each day.
  • Swallow tablets whole; do not chew or crush.
  • In case of vomiting after taking the medication, do not take an additional dose; continue with the regular dosing schedule.
  • The initial low dose increased after 7 days of treatment.

  • If brigatinib is not taken for 14 days or longer, consult with your doctor before restarting.

Common side effects may include nausea, diarrhea, vomiting, tiredness, rash, headache, numbness or pain in hands or feet, joint or back pain, loss of appetite, and difficulty sleeping. Serious side effects may include difficulty breathing, chest pain, persistent cough, fever, severe headache, blurred vision, extreme thirst, frequent urination, muscle pain or weakness, and slow or irregular heartbeat.

Non-small cell Lung Cancer (NSCLC) is indicated for anaplastic lymphoma kinase (ALK)-positive metastatic NSCLC in patients who have progressed on or are intolerant to crizotinib.

  • Brigatinib is a tyrosine kinase inhibitor with in vitro activity against multiple kinases, including ALK, ROS1, IGF-1R, FLT-3, EGFR deletion, and point mutations.
  • Inhibits autophosphorylation of ALK and downstream signaling proteins.
  • Demonstrated antitumor activity against EML4-ALK-positive NSCLC xenografts in mice.

Absorption:

  • Median time to peak concentration (Tmax) ranged from 1 to 4 hours after single oral doses of Brigatinib (30 to 240 mg).

Distribution:

  • Brigatinib is 66% bound to human plasma proteins.
  • The mean apparent volume of distribution (Vz/F) at steady-state is 153 L.

Elimination:

  • The mean apparent oral clearance (CL/F) at steady-state is 12.7 L/h.
  • The mean plasma elimination half-life is 25 hours.

Metabolism:

  • Males with female partners of reproductive potential should use effective contraception during treatment and for at least 3 months after the last dose.
  • Pregnancy and lactation are contraindications.
